The fuel pump runs till the carbs are full then stops and continues as needed. It's a LOW pressure system with Carbs.

Probably the pilot jets being clogged, or a stuck float needle not letting fuel into the bowl. Otherwise it prolly has gotten cold out, and leaned the bike so much you can't take off. So I'd also bet you have a slip on and the pilot adjustors aren't drilled out at all to adjust richer, etc.
On take off, your on the pilot jet circuit on the carbs. That's where your problem will more than likely be. Otherwise, maybe the TPS is all forked up, but it shouldn't advance the ignition that much to where it stalls out...
